Fisetin: Powerful Senotherapeutic Potential*


Fisetin is a bioactive flavonol found in fruits and vegetables such as strawberry, apple, persimmon, grape, onion, and cucumber. Several studies have reported fisetin to have neurotrophic, antioxidant, and anti-inflammatory properties.* [1] Its researched anti-aging activity is thought to come from its ability to target cells that have undergone a process called senescence.* Senescence is when a cell ages and permanently stops dividing but does not die. Over time, these cells build up in tissues throughout the body and can release substances that promote inflammation and affect nearby healthy cells.

Fisetin is an active area of senolytic research. Studies have included work on its senolytic activity alongside the D+Q combination (Dasatinib and Quercetin), and ongoing human research is exploring senolytics in various populations. In a longitudinal study that measured biological age using DNA methylation clocks, researchers examined the effects of D+Q and fisetin on senolytic-related markers.* [2]

A 2018 study [3] set out to test ten flavonoids for senolytic properties. The researchers noted that D+Q is a potent senolytic studied in the context of several age-related conditions, and aimed to identify natural flavonoids with even greater senolytic activity.

The study examined human fibroblasts that had become senescent through oxidative and genotoxic stress, and further tested the flavonoids in mice, evaluating age-related histopathology, disease markers, health span, and lifespan. Of the 10 flavonoids tested, fisetin was the most potent senolytic.* Fisetin reduced senescence markers in multiple tissues, consistent with a hit-and-run senolytic mechanism.* The authors reported that fisetin showed senotherapeutic activity in both human and mouse tissue, and that in mice, fisetin improved tissue homeostasis, reduced age-related pathology, and extended median and maximum lifespan.* Please be aware that findings from animal studies and research on human cell lines may not necessarily reflect outcomes observed in humans.

Fisetin has been studied for its activity on the AMPK, MAPK, and mTOR pathways, and for inhibition of the CDK1 and CDK4 kinase enzymes.* [4] A 2015 study examined how AMPK activation relates to aging [5], while a 2019 review described mTOR's role as a central regulator of lifespan and aging across a wide range of organisms.* [6]

mTOR is a protein kinase that helps regulate cellular metabolism, autophagy, survival, proliferation, and other processes involved in cellular homeostasis. It is the target of the compound Rapamycin, which is noted in the literature as the compound most consistently shown to extend mammalian lifespan. Some studies have explored whether other natural compounds, including fisetin, may also act on mTOR.*
